After a meal your body releases amylin, the hormone that passes on the signal for "enough". Cagrilintide, development code AM833, mimics that hormone and stays active considerably longer. As a research molecule it therefore turns on appetite and weight, and in the literature it almost always stands next to semaglutide, in the combination called CagriSema. The satiety signal, mimicked
What cagrilintide turns on is satiety. It is a long-acting amylin analogue, and amylin is precisely the hormone that produces the full feeling after a meal. The peptide is thought to be of interest for research around weight and metabolism for that reason.
Where in the body does it act?
The point of action sits at the amylin receptors in the brain. The feeling of fullness is thought to be strengthened there and the stomach to empty more slowly, so food stays put for longer. Amylin runs along a different route from GLP-1, and it is exactly that complement that makes it usable in research. Long-acting means one administration lasts longer; studies work on a weekly rhythm. Most of the work still plays out in a research setting.
Why semaglutide is almost always beside it
In nearly every study of cagrilintide, semaglutide turns up. That combination, known as CagriSema, is studied because the two run along different routes: amylin on one side, GLP-1 on the other. In Phase 3 research the combination was linked to stronger effects on weight and blood sugar than either of the two on its own. If you want to place them side by side, semaglutide sits separately in the range. We supply single vials, not a pre-mixed combination.
Complement or all-rounder: the difference from retatrutide
Both are researched around weight, but they are very different molecules. Retatrutide plays three receptors single-handedly, namely GLP-1, GIP and glucagon. Cagrilintide plays the amylin route and is usually studied alongside a GLP-1 such as semaglutide, not as a replacement for it. Making it up and the weekly rhythm
You receive a lyophilised vial that you make up yourself with bacteriostatic water. Because of the long duration of action, research usually administers weekly; the exact protocols vary by study and are documented in the literature. What is reported about tolerability?
What comes up in research sits mainly in the gastrointestinal area and is mild to moderate: nausea, vomiting, diarrhoea or constipation, similar to other substances in this group. On the whole it was well tolerated in the studies. Hard data on use outside of studies is still missing.
