Gonadorelin is the synthetic form of GnRH, the ten-amino-acid hormone with which the hypothalamus prompts the pituitary to release LH and FSH. That places it right at the start of the hormone chain steering the gonads, and it is precisely that position which makes it interesting in research. What gonadorelin research turns on
Gonadorelin is the synthetic form of GnRH (gonadotropin-releasing hormone), the hormone with which the hypothalamus drives the pituitary. In short: gonadorelin signals the pituitary to release two other hormones, LH (luteinizing hormone) and FSH (follicle-stimulating hormone). Those two in turn drive the gonads. In research, gonadorelin is therefore used to study the pituitary-gonadal axis. It was also used as a medicine in fertility research and diagnostics, but those branded products (Factrel, LutrePulse) are largely off the market.
The pulsing signal from the hypothalamus
What is special about gonadorelin lies in the rhythm. The pituitary responds to pulses of GnRH, not to a constant stream. When gonadorelin is given in short pulses in research, it stimulates the release of LH and FSH, it mimics the natural rhythm of the hypothalamus. When it is given continuously, the opposite happens: the receptors become insensitive and LH/FSH release falls. That dual behaviour, stimulating in pulses, suppressing when continuous, is exactly why gonadorelin is of interest in research. Placed next to hCG
Gonadorelin and hCG are often compared because both relate to the sex-hormone axis, but they act at a very different point. Gonadorelin works at the top of the chain: it prompts the pituitary to make LH and FSH itself. hCG skips that step, it mimics LH and acts directly on the gonads, bypassing the pituitary. In the research and TRT context, gonadorelin is therefore seen as the substance that keeps the natural axis switched on, while hCG mimics the axis from below. The axis underneath TRT research
In the context of testosterone replacement (TRT), gonadorelin is a much-discussed topic. The idea behind it: during an external testosterone course, the body-own LH/FSH production stalls, which makes the gonads less active. It is thought that pulsatile GnRH stimulation may keep that axis switched on in research. What is known about this is described in the scientific literature.
What is reported about tolerability?
In the clinical literature, gonadorelin is generally well tolerated. The described side effects are mostly mild: headache, nausea and an irritated or painful injection site; sometimes hot flushes. These are observations from medical use under supervision, not data that says anything about laboratory use. Making it up and storing it

Gonadorelin in solution is less stable than many other peptides: after mixing it goes in the fridge (2–8 °C), protected from light, and is used within a short time; for longer storage the unopened vial can go in the freezer.
