What sets it apart is the number of points it acts on: where most compounds in this class play one or two receptors, this peptide plays three at once, namely GLP-1, GIP and glucagon. Eli Lilly is studying it in a Phase 3 programme; as a medicine it is not yet approved. Why it draws so much talk
Among metabolic research peptides no name comes up more often at the moment, and that traces back to the striking weight results from Eli Lilly's large Phase 3 research. The explanation is sought in the number of switches pressed at once: GLP-1, GIP and glucagon together steer hunger, blood sugar and energy expenditure. That threefold grip is thought to reach further than older compounds such as semaglutide and tirzepatide.
Three pathways side by side
Put simply, three things happen at once. Through GLP-1 and GIP, appetite is curbed and blood sugar kept in balance. On top of that comes the glucagon pathway, which gives energy expenditure a push, and no other known peptide in this class has that third track. The sum is thought to go further than a single GLP-1 compound on its own. The full picture comes from the Phase 3 TRIUMPH programme; approval as a medicine has not been granted.
Placed next to semaglutide and tirzepatide
The distinction fits in one sentence: semaglutide works one receptor, tirzepatide two, retatrutide three. Semaglutide, known under the brand name Ozempic, is a single GLP-1 agonist. Tirzepatide, sold as Mounjaro, adds GIP. Retatrutide lays the glucagon pathway on top of that. So it is not a variant of Ozempic but its own, newer research peptide, and that extra mechanism is exactly why it is studied.
What did the research turn up?
The reactions reported most often in the Phase 3 research sat in the gastrointestinal area: nausea, reduced appetite, sometimes diarrhoea. They were usually mild and appeared mostly in the opening phase, a picture that matches what is known of other GLP-1 compounds.
