It inhibits NNMT, an enzyme that mops up nicotinamide, and nicotinamide is precisely a building block of NAD+. Slow that enzyme and more building material remains, and that is what research into this molecule turns on. What 5-Amino-1MQ research turns on
5-Amino-1MQ is an inhibitor of the enzyme NNMT. In the fat cell, this enzyme is involved in how quickly fat is burned and how large fat cells grow; in research, NNMT stands out mainly because it is elevated in obesity. By dialling NNMT down, animal and cell studies look at whether the cell shifts more towards fat oxidation and whether NAD+ levels rise. It is assumed that this is where the interest in 5-Amino-1MQ comes from in research on metabolism and body composition. We make no health claims about it ourselves, at BasePurity it is purely a research material, not for use in humans.
The enzyme NNMT as the point of action
The enzyme NNMT uses up two things the cell badly needs: a methyl group (from SAM, S-adenosylmethionine) and nicotinamide, the building block the cell makes NAD+ from. By blocking NNMT, 5-Amino-1MQ spares both in research: more nicotinamide is left to recycle into NAD+, and the SAM economy is less burdened. That higher NAD+ availability is also where the link to the sirtuins, the maintenance enzymes of the cell, comes from. Most of what we know about this comes from cell and animal studies.
Why this is not a peptide
This is the biggest misconception around 5-Amino-1MQ: it is often called a "peptide", but it is not one. It is a small synthetic molecule (a quinolinium derivative), small enough to cross the cell membrane on its own, unlike most research peptides. It only gets mentioned in the same breath as peptides because it is studied in the same metabolic and longevity context. For the lab, the difference mostly matters for how you dissolve and store it; the specifications are in the table at the bottom of this page.
The link to the NAD+ supply
The reason 5-Amino-1MQ gets so much attention in the longevity corner is its indirect route to NAD+. NAD+ is the coenzyme cells use to release energy, and its natural supply falls as we age. Because NNMT siphons off nicotinamide that would otherwise become NAD+, research looks at whether inhibiting NNMT helps keep NAD+ levels higher. That puts 5-Amino-1MQ in the same line of research as NAD+ itself, but approaching the problem from the other side. What is known about tolerability?
Because 5-Amino-1MQ is relatively new, almost all knowledge comes from cell and animal studies; large long-term human data are lacking. In those studies the molecule is known for a mild profile, partly because it acts selectively on a single enzyme (NNMT) rather than on the whole body. Exactly what happens with prolonged human use simply has not been investigated yet.
